初步探索了罗非鱼肝脏原代细胞分离、纯化的方法,并在此基础上研究培养基中外源添加不同饱和度的脂肪酸(16:0,18:1 n-9,18:2n-6,20:4 n-6,20:5 n-3和22:6 n-3)对其与脂类代谢相关基因(LPL、MDH和PPARα)mRNA表达的影响.结果表明:60 g,2 min,1次和30 g,2 min,2次的离心条件获得较高细胞产量及纯度;随脂肪酸不饱和程度的增加LPL和PPARαmRNA表达均呈现明显的上升趋势,而MDH则波动性较大.  相似文献

用气相色谱—质谱法分析了7属12株细菌的全细胞酸性水解物,检测出初步化学定名的脂肪酸19种及其它羧酸5种,即7—CH3—C13:0、11—CH3—C13:1^△9-、C—14:O、C14:1^△3、3—CH3—C14:O、3—OH—C14:O、C15:O、14—CH3—C15:O、C16:O、15—CH3—C16:O、C17:O、C17:1^△6、C17:1^△12、16—CH3—C17:O、C18:1^△10、C18:2、C18:3、C19:1^△12、10、11—OH—C20:O、苯二甲酸、苯十二烷酸、苯十四烷酸、喹啉乙酸和喹啉丁酸。供试菌株含量丰富的脂肪酸有C17:1^△6、14—CH3—C15:O、C16:O、15—CH3—C16:O。气杆菌属和变形杆菌属的14—CH3—C15:O含量大于或接近C17:1^△6的含量,而芽孢杆菌、醋杆菌、葡萄球菌、短杆菌和假单胞菌属14—CH3—C15:O的含量少于C17:1^△6的百分含量,各属和各种间的脂肪酸成分都具有一定的特征。本结果为细菌的化学分类和分子细菌学的研究提供资料。  相似文献

采用气相色谱仪测定了小荚蛏不同生理阶段(配子发生期、成熟期和排放期)软体部脂肪酸的组成.结果表明,小荚蛏软体部含有23种脂肪酸,其中饱和脂肪酸(SFA)5种,单不饱和脂肪酸(MUFA)5种,多不饱和脂肪酸(PUFA)13种.各生理阶段的SFA、MUFA和PUFA含量差异极显著,23种脂肪酸除C20:3含量无显著差异外,其余22种脂肪酸含量差异均显著;SFA含量以排放期为最高,配子发生期为最低;MUFA含量在配子发生期高于成熟期和排放期,PUFA含量在配子发生期和成熟期高于排放期.  相似文献

采用生物化学方法的方法,对大鳍鱯繁殖期的性腺、肝脏、肌肉的氨基酸和脂肪酸进行测定.结果表明:大鳍鱯雄性亲鱼精巢、肝脏和肌肉中,肌肉中氨基酸总量最高,为40.90g/100g;其次是精巢,为26.38g/100g;肝脏中含量最低,为18.38g/100g。从必需氨基酸占总氨基酸的比例来看,精巢最高,占53.41%;其次是肌肉,占52.93%;肝脏中最低,占51.96%。雌性亲鱼肌肉中氨基酸总量最高,为40.61g/100g;其次是卵巢,为22.42g/100g;肝脏中含量最低,为17.04g/100g。从必需氨基酸占总氨基酸的比例来看,卵巢最高,占52.85%;其次是肝脏,占51.76%;肌肉中最低,占49.OO%。从脂肪酸组成来看,雌雄亲鱼均为MUFA所占比例最高,其次是SFA,PUFA比例最小。雄鱼与雌鱼的三种组织器官中,所占比例较高的脂肪酸相似,它们是C18:1n.9(油酸)、C16:0(软脂酸)、C18:2n-6(亚油酸)和C16:1n-7(棕榈酸)。研究揭示了大鳍鱯繁殖期雌、雄亲鱼主要组织器官在物质构成上的特性。  相似文献

以Sr(OOCCH3)2·H2O,Ti(OC4H9)4·Nb(C2H5O)5为原料,用溶胶-凝胶工艺成功地进行了SrNb0.05Ti0.95O3(SNTO)膜的制备,溶胶薄膜经过575℃-725℃/60min退火形成立方钙钛矿结构。用XRD、SEM等进行了结构和形貌表征,证明了该薄膜是纳米晶体结构,制定了SNTO薄膜的最佳退火工艺。SNTO薄膜的介电特性分析:掺杂Nb^5+能使SrTiO3转化为n-半导体。  相似文献

采用索式提取法对螺旋藻中的脂肪油进行提取,并对脂肪酸进行甲酯化处理,利用气相色谱-质谱联用仪进行分析测定.螺旋藻共检测出6种脂肪酸,不饱和脂肪酸为4种,相对百分含量占检出所有脂肪酸的57.44%,其中,二十碳五烯酸(EPA)为12.17%,亚油酸为7.25%,5,8,11,14-二十碳四烯酸(花生四烯酸)为22.12%.  相似文献

目的:获得一株高产雄甾-4-烯-3,17-二酮( AD )的Mycobacterium neoaurum突变株。
  创新点:获得了一株3-甾酮-Δ1-脱氢酶(KSDD)酶活缺陷型的高产 AD的诱变菌株 Mycobacterium neo-aurum ZADF-4,并采用菌落显色法筛选 KSDD酶活缺陷型M. neoaurum突变株。
  方法:(1)诱变方法:采用常压室温等离子体(ARTP)诱变技术来处理出发菌株 M. neoaurum ZAD。ARTP 诱变条件如下:功率40 W ,气流量12.5 L/min,辐射距离1 cm,样品体积10μl,辐射时间为60、90、120、150和180 s;致死率统计优化后,最适辐射时间为150 s,致死率为90%~96%。(2)筛选方法:将ARTP诱变处理后的菌株点种在硝酸纤维滤膜上,30°C培养2 d,然后将长有菌落的滤膜小心取出并漂浮在4 mg/ml二氯靛酚(DCPIP)溶液(0.1 mmol/L磷酸缓冲液pH 7.0),30°C培养1 d直到全部菌落染成蓝色。然后将该滤膜取出,漂浮在250 mmol/L AD溶液(2%甲醇和50 mmol/L Tris pH 7.0缓冲液),室温放置15 min左右,观察菌落颜色变化。KSDD在底物 AD 存在时会脱氢产生雄甾-1,4-二烯-3,17-二酮(ADD)和H+,H+可以使被DCPIP染成蓝色的菌株褪色。因此,酶活缺陷型的菌株会仍保持蓝色,而酶活高的菌株会褪色为黄色(图3)。(3)对获得的潜在的高产AD菌株进行进一步的酶活检测以及产量验证,以期获得最优的突变株。
  结论:获得了4株具有潜在的高产AD能力的菌株,其中,最优的突变株ZADF-4的KSDD酶活相较于出发菌株ZAD下降了81.2%(图4),活性胶也证明其KSDD酶活相较于出发菌株下降明显(图5)。薄层色谱法(TLC)和高效液相色谱法(HPLC)实验证明突变株 ZADF-4中,AD的产量有了明显的提高(图6和图7),提高到了(6.28±0.11) g/L, AD/ADD提高到8:1,AD的摩尔产率达到60.3%(表1)。对出发菌株ZAD和突变株ZADF-4的ksdd基因进行克隆和序列比对,发现ZADF-4的ksdd序列在5'端缺失9个核苷酸(atgttctac),导致3个氨基酸(MFY)的缺失;还发生了两个点突变,其中一个是无义突变(g.15a>6t),另一个是有义突变(g.413c>404t),并引起了相应位置上的氨基酸变化(p.138S>135L)。上述的基因突变及其引起的氨基酸序列的变化可能是引起M. neoaurum ZADF-4中KSDD酶活降低及AD产量提高的主要原因。  相似文献

采用同时蒸馏萃取方法对松乳菇挥发性成分进行提取,并用气相色谱—质谱法对提取物进行化学成分分析.从同时蒸馏萃取提取物中鉴定出了67个成分,并以归一化法测定了各个成分的质量分数.其中超过2%的有9种:1-辛烯-3-醇(2.27%),丁酸丁酯(13.52%),(E)-2-辛烯醛(2.88%),n-癸酸(4.99%),[1 aR-(1 a..α,4..β,4 a..β,7..β,7 a..β,7b..α)]-十氢-1,1,4,7-四甲基-1H-环丙烯并[e]薁(4.16%),n-十六烷酸(4.75%),(Z)-9,17-十八碳二烯醛(5.19%),(E)-9-十八碳烯酸(3.33%),十八烷酸(2.13%).  相似文献

以葡萄藻为原料,通过单因素和正交试验,探讨了料液比、微波温度、微波时间、微波功率对提取粗脂肪的影响。实验结果表明最佳的工艺条件为:微波功率为400W,微波温度60℃,微波时间120s,料液比1∶20。此条件下重复实验,粗脂肪得率为15.08%。对葡萄藻脂肪酸成分分析,主要脂肪酸成分为C16∶0,C16∶1(n-7),C18∶1(n-9),C20∶5(n-3)。其中C16∶1(n-7)的含量达到37.68%。无二十二碳六烯酸(DHA)含量。  相似文献

We examined the effects of conjugated linoleic acid (CLA) on growth, fatty acid composition and enzyme activity of fatty acid oxidation in the liver of large yellow croaker. We divided 1600 fish (average initial weight 150 g) into 4 groups and reared them in 8 cages. Four dietary treatments were formulated to contain 0%, 1%, 2% and 4% (w/w) CLA, respectively. The fish were fed for 10 weeks ad libitum twice daily. We found that the dietary CLA had no effect on growth, biometric parameters and whole body proximate (P>0.05), but showed some significant effects on the fatty acid composition in both muscle and the liver.The activities oflipogenic enzymes were slightly depressed in fish fed with increasing levels of CLA when compared with control (P>0.05). Dietary CLA supplementation had no effects on liver lipid content, but significantly increased the contents of saturated fatty acids (SFA) and polyunsaturated fatty acids (PUFA) (P<0.05) and decreased monounsaturated fatty acid (MUFA) content in both muscle and the liver. Dietary CLA inclusion resulted in significant increases of the biologically active cis-9, trans-11 and trans-10, cis-12 isomers in both tissues (P<0.05). The total accumulation of CLA was higher in the liver (3.83%, w/w) than in muscle (3.77%, w/w) when fed with 4% (w/w) CLA. This study demonstrates that large yellow croakers are capable of absorbing and depositing CLA and long-chain n-3 PUFA in the liver and muscle, showing that this species fed with CLA could be an important human food source for these healthful fatty acids.  相似文献

As the consumption of fructose and saturated fatty acids (FAs) has greatly increased in western diets and is linked with an increased risk of metabolic syndrome, the aim of this study was to investigate the effects of a moderate (10 weeks) and a prolonged (30 weeks) high fructose and saturated fatty acid (HFS) diet on plasma FA composition in rats. The effects of a few weeks of HFS diet had already been described, but in this paper we tried to establish whether these effects persist or if they are modified after 10 or 30 weeks. We hypothesized that the plasma FA profile would be altered between 10 and 30 weeks of the HFS diet. Rats fed with either the HFS or a standard diet were tested after 10 weeks and again after 30 weeks. After 10 weeks of feeding, HFS-fed rats developed the metabolic syndrome, as manifested by an increase in fasting insulinemia, total cholesterol and triglyceride levels, as well as by impaired glucose tolerance. Furthermore, the plasma FA profile of the HFS group showed higher proportions of monounsaturated FAs like palmitoleic acid [16:1(n-7)] and oleic acid [18:1(n-9)], whereas the proportions of some polyunsaturated n-6 FAs, such as linoleic acid [18:2(n-6)] and arachidonic acid [20:4(n-6)], were lower than those in the control group. After 30 weeks of the HFS diet, we observed changes mainly in the levels of 16:1(n-7) (decreased) and 20:4(n-6) (increased). Together, our results suggest that an HFS diet could lead to an adaptive response of the plasma FA profile over time, in association with the development of the metabolic syndrome.  相似文献

以黄瓜品种"津研4号"为材料,研究了低温下黄瓜种子萌发过程中脂肪酶活性和脂肪酸含量的变化.结果表明:常温下黄瓜种子萌发过程中脂肪酶活性随时间呈现先升高再降低的变化趋势,脂肪酸含量随时间增加而增加;低温胁迫下脂肪酶活性、脂肪酸含量也呈现出与常温相同的趋势,但脂肪酶活性高峰出现的时间远远滞后于常温,脂肪酶的最大活性高于常温.  相似文献

Polyunsaturated fatty acids (PUFAs) possess anti-cancer action both in vitro and in vivo. In the present study, we detected cell viability with methyl thiazolyl tetrazolium (MTT) assay and cell membrane permeability with propidium iodide (PI) fluorescence dyeing, and calculated cell membrane fluidity change as fluorescence anisotropy. Fatty acid content in cells was measured by gas chromatography/mass spectroscopy (GC/MS), and the relationship between fatty acid composition and cell viability was studied. We observed that n-6 PUFA linoleic acid (LA) inhibited tumor cell growth at high concentrations (≥300 μmol/L), while low concentrations (100–200 μmol/L) seemed to promote cell proliferation. Analyses of cell membrane permeability, cell membrane fluidity, and cell fatty acid composition suggested that the anti-cancer action of LA could be related to changes in the ratio of n-6 to n-3 PUFAs. We observed that pre-incubation of cancer cells with 100 μmol/L LA for 24 h enhanced cell sensitivity to the cytotoxic action of LA, whereas undifferentiated cell line LoVo seemed to have a distinct path in LA-induced death. These results showed that one of the mechanisms by which supplementation of LA induces cancer cell death could be altering the ratio of n-6/n-3 PUFAs, and this may be related to cell differentiation status.  相似文献
