从土壤中分离、筛选到一株产碱性磷酸酶的菌株HB1,并且该菌株产生的碱性磷酸酶具有较好的热稳定性.根据形态学观察、生理生化特征,DNA中(G+C)mol%测定和16S rDNA序列同源性分析表明该菌为Bacillus licheniformi(地衣芽孢杆菌).调节基础培养基起始pH为7.5,添加Mn2+100 mM/L,发酵24 h,可使HB1菌株细胞内外酶活分别达到5.3 U/mL和5.9 U/mL.  相似文献

介绍了150株三元能量处理妥布拉霉素(TOB)正突变株的筛选研究,通过初筛获得25株TBO突变株,从中分离出15株高产菌,进一步复筛双从中挑选出5株产量更高的产高菌株。通过最佳培养基配方的发酵试验,确定联抗性能最佳的菌株为QB-130-S1。实验表明:三元能量用于TOB菌诱变,不仅提高了产抗水平,而且菌株生长时间缩短,经4次传代,高产菌株的遗传性能稳定。  相似文献

对黑曲霉HYA4菌株所产果胶酶的酶学性质进行了研究。结果表明:该酶反应最适温度为48℃,果胶酶最适pH值为4.8,果胶酶活力稳定的pH范围为3.0-6.0;Km值为4.33mg/mL;金属离子Ca2+、Mg2+对果胶酶有较强的激活作用,Ba2+对果胶酶也有一定的激活作用,Ag+、Mn2+、Zn2+、Fe3+、K+对果胶酶有抑制作用。由SDS-PAGE测定果胶酶的相对分子量为46000±3000Da;IEF电泳测定其等电点为4.7±0.2。  相似文献

对一株产纤维素酶的黄曲霉进行发酵条件优化和酶学性质初步探究,利用DNS法测定CMC酶活、滤纸酶活,结果显示此菌株纤维素酶最佳反应温度为50℃,最佳反应pH为7.0。在金属离子中,发现Zn~(2+)具有抑制作用,最大抑制浓度为0.4 mg/m L,Mg~(2+)、Mn~(2+)、Ca~(2+)、Fe~(3+)、Fe~(2+)具有促进作用,最适浓度分别为0.4、1、1、0.8、1.4 mg/mL。蛋白胨和酵母粉具有促进作用,最适浓度分别为1.0%、0.6%,最适pH为6.0。综合优化后,测得滤纸酶活和CMC酶活分别为7.73 U/mL、11.28 U/mL,相比优化前提高了38.8%和43.7%。  相似文献

从患病鲫、鲢、鳙、鳊等鱼体上分离出一菌株,经腹腔注射及浸泡感染,能引起鱼患典型的出血病症和死亡,其半数致死量(LD_(50))为1.48×10~5个菌/鱼.该菌发酵分解糖类,氧化酶和过氧化氢酶实验为阳性,具极生单鞭毛,革兰氏阴性,对弧菌抑制剂不敏感,经生理生化特性测定及形态、培养特征观察,确认为嗜水气单胞菌(Aermonas hydrophila).并测定了该菌对抗生素的敏感性及生长特性。  相似文献

本文报道使用三株霍乱弧菌型特异性单克隆抗体(VC-McAb),以干菌快速酶标法对1088株历年分离保存的霍乱弧菌EL-Tor生物型菌株进行了血清型和流行菌型的分型鉴定,并与原有的血清学、噬菌体和生物分型比较,证明VC-McAb对菌株的分型率分别为小川型流行株90.42%,小川型非流行株97.04%,稻叶型流行株89.34%和稻叶型非流行株93.94%。本法快速(仅需2h)、简便、敏感、特异,对霍乱弧菌血清型和流行菌型的快速分型鉴定,具有较大的实用价值。  相似文献

从岱山盐田采集土壤样品,利用含不同Na Cl浓度的RM培养基分离中度嗜盐菌,并采用抑菌圈法进行抗菌活性筛选。结果共分离得到106株中度嗜盐细菌菌株,其中有27株显示出对革兰氏阳性菌、革兰氏阴性菌和人类病原真菌的有效抗性。通过基于16S r RNA基因序列的系统发育分析,具有抗菌活性的27株菌株分别属于7个属,即嗜盐单胞菌属(Halomonas)、喜盐芽孢杆菌属(Halobacillus)、海杆菌属(Marinobacter)、产微球茎菌属(Microbulbifer)、Piscibacillus属、芽孢杆菌属(Bacillus)和枝芽孢杆菌属(Virgibacillus)。研究结果表明,岱山盐田土壤中存在较丰富的中度嗜盐菌种类,是筛选抗菌活性菌株的潜在资源库。  相似文献

Bats, the only mammals known to possess the real ability to fly, have fascinated zoologists for a long time with their long-distance migration. How do they find their way home from an altitude of kilometers above sea level, where their stunt of ultrasonic echolocation does not work? Recent discoveries supported the theory that bats might be able to use magnetic clues to figure out their own position,just like what birds do. However, the latest results from a joint study by a group of researchers from the CAS, East China Normal University and University of Auckland reveal that bats apply an interesting strategy different from the one underlying birds' orientation technique, though they both are believed to derive information about their position from the subtle changes in the magnetic field of the Earth.  相似文献
